For synthesis of nucleotide-modified mRNA, unmodified uridine triphosphate was fully replaced with chemically modified nucleotides (Ψ, m1Ψ, 5moU; 100 mM; all Jena Bioscience, Germany). mRNA integrity was verified by running 2 uL of mRNA with 5 uL loading dye (included in the IVT kit) on a 0.9% RNAse-free agarose gel. mRNA concentration was adjusted to 1 mg mL−1 using UV–vis-spectroscopy (NanoDrop 1000; Peqlab, Erlangen, Germany).
