Preparation: VZV infected cells are harvested and virus is extracted by alkaline treatment and sonication. The preparation is purified by ultracentrifugation through a sucrose cushion. The resulting antigen consists of a high concentration of virus and viral components, it may contain residual host cell material. The antigen is suspended in 100 mM glycine buffer, pH 9.5.

Inactivation: Physical inactivation. Inactivity is confirmed by attempted growth under original culture conditions. Since no procedure can guarantee absolute sterility, the reagent should be handled with appropriate safety precautions.
