Alkyne Agarose is an efficient matrix to covalently capture CLICK-functionalized proteins by a Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC). The proteins of interest need to be metabolically, enzymatically or chemically azide-tagged. Subsequently, the alkyne agarose containing the covalently attached proteins can be washed with high stringency, virtually eliminating any non-specifically bound proteins.
