The peptide RRRDDDSDDD can be used as a substrate for CK2 (casein kinase 2) in in vitro kinase assays. It is phosphorylated by CK2 with a Km of 50 μM and a Vmax of 159.7 nmol/min/mg.
Casein kinase 2 (CK2) is a ubiquitous pleiotropic proliferation-associated serine/threonine protein kinase. Similar to protein kinase A, CK2 is a tetramer made up from two different subunits a2b2. The catalytic a-subunit corresponds to the C-subunit of PKA, the non-catalytic b-subunit is unique and differs from the R-subunit of PKA in all known features.
Recently it was shown that the b subunit is trifunctional: (i) it confers stability to the holoenzyme, (ii) it increases enzyme activity, and (iii) it determines substrate specificity.
