Featured Article March 2026 - Antiviral nucleotide analogs

"Allosteric targeting with antiviral nucleotide analogs allows fine tuning of SAMHD1 dNTPase activity." by Dirks et al.[1] (Journal of Biological Chemistry, Jan 29, 2026) is our featured scientific article for March.

In a recent publication, Dirks et al. explore how antiviral nucleotide analogs can influence the activity of the cellular enzyme SAMHD1.

SAMHD1 plays a central role in regulating intracellular dNTP pools by hydrolyzing deoxynucleoside triphosphates. Through this mechanism, the enzyme acts as an important antiviral defense factor, limiting the availability of nucleotides required for viral DNA synthesis.

In this study, the authors demonstrate that certain antiviral nucleotide analogs can bind to regulatory sites of SAMHD1 and modulate its enzymatic activity through allosteric mechanisms. Rather than directly inhibiting the catalytic center, these molecules influence how the enzyme assembles and becomes activated, thereby allowing a more subtle control of dNTP hydrolysis.

Why does this matter?

  • Antiviral nucleotide analogs can modulate host nucleotide metabolism, not just viral replication
  • SAMHD1 activity can be fine-tuned via allosteric regulation rather than simply inhibited
  • Enriches understanding of a more precise way to control intracellular dNTP levels
  • Opens new possibilities for next-generation antiviral drug design targeting both host and virus
