RNA synthesis monitoring (potentially)

5-AmU (5-Azidomethyl-uridine) can potentially be used as a replacement for BrU (5-Bromo-uridine) or the copper-catalyst requiring 5-EU (5-Ethynyl-uridine) to measure de novo RNA synthesis in proliferating cells. 5-AmU is cell permeable and might incorporate into nascent RNA instead of its natural analog uridine. This assumption is based on the previously demonstrated suitability of 5-AmdU (5-Azidomethyl-2'-deoxyuridine) for DNA synthesis monitoring[1] as well as 5-AmUTP (5-Azidomethyl-UTP) for RNA synthesis monitoring[2].

The resulting azide-functionalized RNA can subsequently be detected via Cu(I)-catalyzed (CuAAC) or Cu(I)-free strain promoted (SPAAC) Click Chemistry that offers the choice to

  • introduce a Biotin group for subsequent purification tasks (via Alkynes of Biotin or DBCO-functionalized Biotin, respectivly)
  • introduce a fluorescent group for subsequent microscopic imaging (via Alkynes of fluorescent dyes or DBCO-functionalized fluorescent dyes, respectivly).
