1) Biotin-18-dUTP is a common replacement for Biotin-16-dUTP.
The ability of the common DNA polymerases to incorporate biotinylated nucleotides allows construction of biotinylated double-stranded and/or single-stranded DNA. This labeled DNA is routinely applied for monitoring of gene expression, in situ hybridization, and four-color DNA sequencing. Furthermore, the high affinity of biotin to avidin/streptavidin allows isolation of hybrid molecules (biotinylated DNA in complex with a ligand) by affinity chromatography.
