Affinity chromatography is a widely used method for protein purification. It utilizes the affinity of a "tag" attached to the protein of interest to bind to a specific ligand that is immobilized by a covalent bond to a chromatographic bed material (matrix).
Crude cell-lysate samples are loaded onto the affinity column under conditions that ensure the maximum specific binding of the tagged protein to the immobilized ligand. Other proteins that do not contain the "tag" do not bind the immobilized ligand and are washed through. Elution of the bound protein can be achieved by changing the experimental conditions to favour desorption.
