The novel labeling kits presented here are designed for site-directed labeling of

  • any protein’s C-terminus using convenient, fast, mild and biocompatible oxyamine–ketone ligation (scheme),
  • an N-terminal cysteine using native chemical ligation with a fluorophore-thioester, or
  • a protein’s C & N-termini simultaneously with two colors in a One-Pot Dual Color labeling reaction for fluorescence resonance energy transfer (FRET) studies

Cloning of the target gene into a modified vector followed by recombinant expression results in a N- or C-terminal modified protein preparation that is subsequently labeled with fluorescent dyes.
 



General method for protein modification at the C terminus: Under mild and biocompatible conditions a protein thioester is transformed with 1,2-bis(oxyamino)ethane into an oxyamino-modified species that in a second step reacts rapidly with ketones carrying a fluorescent label.
