High-level inducible protein expression in LEXSY

  
The new pLEXSY_I-2 vectors, available with selection markers ble or neo allow the regulated expression of proteins either in the cytoplasm or via the secretory pathway. For cytosolic expression (c) BglII, NcoI, or SlaI restriction sites may be used at the 5'-end. Cloning into the SalI or XbaI sites yields proteins fused to a signal peptide (SP) which mediates efficient secretion to the medium (s). At the 3'-end the ORF may be fused to a His6 stretch (H6) via cloning into MspC1, or KpnI sites, whereas insertion into the NotI site will replace the His6 stretch. In both cases a 1 kbp stuffer is replaced by the target ORF.
